Matron's Den is an underground area in Act 5.
Matron's Den is one of the three areas of 
The Pandemonium Event, in Hell only. Its boss, Lilith, drops one of the organs that open Uber Tristram.

Horadric Cube in Harrogath, in Hell:

Key of Destruction, dropped by Nihlathak in Halls of Vaught
The portal leads to Matron's Den, the Forgotten Sands or the Furnace of Pain at random; each opens once per game.
